Population Pharmacokinetics of Labetalol in Hypertensive Patients Using NONMEM

The population pharmacokinetics of labetalol was evaluated based on a mixed-effect model using the NONMEM program. The study was conducted in 58 hypertensive patients, receiving labetalol along with other medications by oral were analyzed according to a two-compartment model in FOCI; method. Labetalol in human plasma was measured by a reverse phase high-performance liquid chromatographic method using UV detection. Additive proportional error model was employed to describe residual and inter-individual variability. The Influence of covariates such as age, gender, bodyweight on to I body clearance, and volume of distribution were examined. Bodyweight showed a significant influence on clearance whereas the volume of distribution was influenced by age. The present population pharmacokinetic model was well described on the individual exposure to labetalol and might have a positive Impact on the management of labetalol therapy in the hypertensive patients.
