SEGREGATION OF MENDELIAN MARKERS IN DOUBLED HAPLOID AND F2-PROGENY OF A BARLEY CROSS

Forty-two doubled haploid (DH) barley lines, derived from the F1 of a cross between a multiple marker stock (MMS) and the cultivar 'Apex' by the Hordeum bulbosum method, were tested for their segregation at 23 Mendelian marker loci. Protein, restriction fragment length polymorphism (RFLP), polymerase chain reaction (PCR), and morphological markers were assayed. DH data were compared with data on 100 F2 plants from the same cross. Chi-square values showed excellent fit to hypothesized ratios for all 23 markers in the DH population. No preferential transmission of alleles from one parent was exhibited. DH lines from the cross MMS X 'Apex' were derived from a random sample of gametes. DH lines should provide a useful vehicle for gene mapping in barley.
