EFFECTS OF BILE-DUCT LIGATION ON SERUM AND HEPATIC 5'-NUCLEOTIDASE ACTIVITIES IN ETHANOL INTOXICATED RATS

摘要
The activity of 5'-nucleotidase (5'-NT), one of the marker enzymes of cholestasis, was measured in rats with acute ethanol intoxication after common bile duct (CBD) ligation and in rats with CBD ligation after chronic ethanol intoxication to investigate the adverse effects of alcohol ingestion to the liver under the hepatobiliary disease. Serum 5'-NT activity in CBD ligated rats combined with acute and chronic alcohol intoxication showed a greater increase than that in CBD ligation alone or that in alcohol intoxication alone. Liver cytosolic 5'-NT activity in CBD ligated rats combined with alcohol intoxication was lower than that in CBD ligation alone. These results indicate that the leak of cytosolic 5'-NT into the serum is facilitated in CBD ligated rats combined with alcohol intoxication than in CBD ligation alone or alcohol intoxication alone due to aggravated hepatic membrane damages. Microsomal 5'-NT was induced by CBD ligation and by alcohol intoxication, and when alcohol intoxication was combined with CBD ligation the induction of microsomal 5'-NT was promoted. According to these results, if alcohol intoxication is combined with cholestasis, the hepatic damage is aggravated.
