Crown ether activates cholesterol oxidase in low water media

Kinetic studies of cholesterol oxidase-catalysed oxidation of cholesterol in water/2-propanol mixtures showed a decrease of Vmax/Km values on the increase of concentration of the organic co-solvent. Addition of 18-crown-6 to the reaction medium results in an increase of Vmax up to 16 times, and Vmax/Km up to 8.4 times, enhancing the activity of cholesterol oxidase in 2-propanol/water (88:12 v/v) to 3.5 times compared to the level observed in 46% 2-propanol.
