In the Republic of Moldova physicians prescribe regularly extemporaneously compounded medicines for children in different pharmaceutical forms. A special role in the efficiency of pediatric pharmacotherapy which involves compounded medications refers to rational use of medicinal substances in association with biopharmaceutical factors regarding formulation. The paper reflects the study of extemporaneous pharmaceutical prescriptions, received and prepared at the Production Department of "Vasile Procopisin" University Pharmaceutical Center (VPUPhC) of "Nicolae Testemitanu" State University of Medicines and Pharmacy (SUMPh) and aims to optimize medical prescriptions and preparation technology. Prescriptions analysis have shown that, extemporaneous prescriptions of semi-solid pharmaceutical form as suppositories, indicated for children, for the last years, have been significantly increased. The rectal route is used not only for a local effect but also for a general-purpose action. It was found that from the total amount of prescriptions contained suppositories - 41,8% are indicated for antibacterial effect, 31% for spasmolytic affect, 20% for anti-inflammatory and antipyretic actions and 7,2% for laxative effect. Suppositories with ampicillin and cefazolin sodium, which founded to be the most prescribed antibacterial substances, were subjected to evaluation in order to optimize their compounding technology. In the result of performed quality essays, it was revealed that rectal preparations of ampicillin prepared on cocoa butter and rectal preparations of cefazolin sodium prepared on semi-synthetic glyceride base, have the highest availability.
