Development of 30 SNP markers for Amorphophallus yunnanensis based on RAD sequencing

摘要
Amorphophallus yunnanensis is a tuber-producing plant species distributed in Southeastern Asia. Wild populations of A. yunnanensis are threatened as a result of human activity. To facilitate future work on the genetics of A. yunnanensis, restriction-site associated DNA sequencing (RAD-seq) was carried out for 28 individuals to identify nuclear single nucleotide polymorphisms (SNPs). Twenty-five primer pairs comprising 30 SNPs were developed and further validated using Sanger sequencing. The observed and expected heterozygosity of each locus varied from 0.0000 to 0.2857, and 0.1913 to 0.4638, respectively. Five loci showed significant departures from Hardy-Weinberg equilibrium. SNP development will enable future population genetics studies, aiding in conservation efforts for this threatened species.
