Paraoxonase L55M polymorphism and coronary artery disease in farmers - A preliminary report

Paraoxonase gene (PON1) codes for an HDL-bound enzyme with a putative role in protecting LDL against peroxidation. In our ongoing case-control study, we have observed an association between CAD and PONI L55M polymorphism in Iranian population. Interestingly, when CAD cases are divided into two non-farmer and farmer subgroups, this association is strikingly accentuated in the subgroup of farmer CAD patients, becoming extremely significant. While in the non-farmer subgroup of CAD patients it dwindles appreciably. Statistical analysis reveals a significant difference in the frequency of L and M alleles between farmer and non-farmer CAD patients. PON1 is known to catalyze the hydrolysis of xenobiotic pesticides, notably organophosphates. On this basis, a possible gene-environment interaction. is suggested.
