HYPOXIA-INDUCIBLE FACTOR-1A PROMOTES THE THERAPEUTIC EFFECT OF ASTRAGALOSIDE IV ON HYPERTENSION

摘要
To explore the effect of HIF-1 alpha on the cardiovascular protective effect of astragaloside IV. Mice were divided into four groups: hypertension model group (LC), astragaloside IV treatment group (LR), HIF-1 alpha overexpression combined with astragaloside IV treatment group (LO), and HIF-1 alpha knockdown combined with astragaloside IV treatment group (LK). The mean blood pressure of the mice was mea-sured on days 1, 7, 14, and 21, and then aortic tissue samples were collected. The expression of HIF-1 alpha-related signaling pathway molecules was detected using western blotting and qPCR, and the concentra-tion of vascular protective molecules was detected using the ELISA method. The mean blood pressure in the LO group was significantly decreased, the JAK/STAT3/HIF-1 alpha signaling pathway was significantly activated, and antiapoptotic and vascular protective molecules were up-regulated. In addition, the concen-tration of vascular protective molecules was significantly increased in the blood samples. Hence, Over -expression of HIF-1 alpha significantly activated the JAK/STAT3 signaling pathway, increased the expression and concentration of vascular protection-related molecules, and enhanced the cardiovascular protective effect of astragaloside IV.
