Drug-in-CD-in-liposome(DCL) is a new drug delivery system that encapsulates cyclodextrin inclusion complexes into liposomes, so it combines the advantages of the two drug delivery systems. DCL can effectively avoid the leakage and rapid release of lipophilic drugs incorporated in the membrane bilayers, which is beneficial to maintain the integrity of liposomes. The dual protection of cyclodextrin and liposome can drastically improve the stability of light-sensitive and heat-sensitive drugs. It allows better control of the release behaviors of drugs and improves the therapeutic effect of drugs. DCL has a wide range of applicable drugs and shows promising properties for applications. This paper reviews the classification, preparation methods, characteristics, and application progress of DCL in drug delivery, which provides methods and references for developing DCLs as a drug delivery system. © 2022 Shanghai Institute of Pharmaceutical Industry.
