Effects of chronic administration of nicotine on stress-induced changes in rats

Effects of 2-week nicotine (Nic) administration on stress-induced changes in rats were studied using 4 groups: Nic+stress (Nic/st), untreated+stress (Un/st), Nic +control (Nic/con) and untreated+control (Un/con). The 17 hr-water-immersion stress induced gastric hemorrhage in Un/st but subtle lesion in Nic/st rats. Hypothalamic adrenaline(Ad) concentration was decreased in Un/st rats, but not in Nic/st or Nic/con rats. Serum ACTH concentration was increased in Un/st rats, but not in Nic/st or Nic/con groups. In contrast, the serum corticosterone, Ad and noradrenaline concentrations were increased in both Un/st and Nic/st rats. Vagotomy abolished the stress-induced gastric hemorrhage completely. Inhibitory effects of Nic on stress-induced gastric hemorrhage may be correlated with modification of hypothalamic Ad concentration. Vagus system plays an important role, while peripheral endocrine systems may contribute little to the inhibition.
