摘要
GdmCl (guanidine hydrochloride)-induced unfolding and refolding of porcine pancreas trypsin have been studied by various physicochemical methods including near and far UV CD ( Circular dichroism) spectrum, intrinsic fluorescence spectroscopy (intensity, emission maximum wavelength, tryptophans anisotropy etc.) as well as ANS (8 - anilino - 1 - naphthalene-sulfonic acid) fluorescence. In our present study, the formation of two unfolding intermediates has been established. They were further confirmed by 'phase diagram' approach. Descriptions of protein unfolding-refolding pathway and the intermediates conformations have also been given.
