A hepatitis C electrochemical immunosensor based on thionine covalently bound to self-assembled monolayer

摘要
A new technique on the preparation of Hepatitis C virus (HCV) electrochemical immunosensor was reported. 3-Mercaptopropionic, acid was assembled on gold electrode to form a self-assemble monolayer. After the modified electrode was activated by 1-[3-(dimethylamino) propyl]-3-ethyl-carbodiimide methyiodide (EDC) and N-hydroxysuccinimide ( NHS), thionine and the HCV horseradish peroxidase (HRP) antibody were covalently bounded to the self-assemble monolayer competitively to form the Hepatitis C electrochemical immunosensor. Cyclic voltammetry and linear scan voltammetry were used to investigate the assembling process and the chemical characteristics of the immunosensor. The linearity of Hepatitis C in the range of 3.2 similar to 16 mg/L with the correlation coefficient of 0.995 was obtained by linear scan voltammetry. While used in the clinical application of human serum, this method was in good agreement with the enzyme linked immunosorbent assay.
