Dissolution profiles in Bio-Dis of Ketoprofen Pellets obtained by Extrusion-spheronization and Fluid Bed Coating. A 2(2) factorial design was used to elucidate the effects of granulometry and coating weight gain on the drug release profiles, in Bio-Dis, of ketoprofen sustained release pellets obtained by extrusion-spheronization and fluid bed coating with Kollicoat (R) EMM 30D. The dissolution profiles were compared using f1/f2, "one-way" ANOVA and Weibull method. The influence of the pH value on drug release was evaluated performing the linear regression of the dissolution curve segments relative to the pH 6.0, 6.8 and 7.2 values. The weight gain has governed the release from the formulations, while the effect of granulometry has just been observed for those with the highest weight gain. The increase on the pH value rose the velocity of ketoprofen release because of the pH-dependent solubility of the drug.
