摘要
In this paper, the effect of injured pancreatic extract and cell growth factors on the induced differentiation of rat adipose mesenchymal stem cells to islet-like cells was investigated. The rat adipose mesenchymal stem cells were extracted and separated from adipose tissues, and were induced to differentiate by injury pancreatic extract and a variety of cytokines respectively or both of two. Insulin-positive cells were identified by dithizone staining, ELISA assay was used to detect the level of insulin expression, and the related gene expression was detected by real-time quantitative polymerase chain reaction (RT-PCR) and immunofluorescence staining. Compared with control group, the number of insulin-positive cells in the experiment groups was increased. The expressions of insulin, PDX-1, insulin in the experiment groups were significantly higher than those in the control group. The combined group more effectively induced ADMS cells into islet-like cells compared with other experimental groups, the difference was statistically significant (P < 0.05). Injured pancreatic extract combining with traditional induction can promote the adipose mesenchymal stem cells to differentiate to islet-like cells more efficiently.
