Comparative analysis of enantioselective separation of novel PPAR agonists by HPLC on cellulose- and amylose-based chiral stationary phases

摘要
Chiral chromatographic resolution of a series of dual PPAR alpha/gamma agonists was performed on the commercially available Chiralcel OD and Chiralpak AD columns in normal phase mode using a mobile phase consisting of a mixture of n-hexane, 2-propanol and trifluoroacetic acid at constant volume ratio. The selected CSPs resolved most compounds as underivatized acids without requiring time consuming analysis. Both electronic features and steric hindrance affected chiral interactions between the analytes and the CSPs. Additional information on the chiral recognition mechanism was deduced from the chromatographic behaviour of some selected methyl esters.
