Sequence Analysis of Wheat WIS2-1A Reverse Transcriptase Response to Low-energy N+ Implantation and 60Coγ-ray Irradiation

摘要
In this paper, retrotransposons WIS2-1A reverse transcriptase conserved sequence of wheat irradiated by different doses of low-energy N+ and Co-60 gamma-ray was studied. The results showed that compared with the standard sequence in the Genebank, sequence length and the homology of WIS2-1A reverse transcriptase were changed, in which low-energy N+ implantation treatment changed greatly. The base mutation types of WIS2-1A reverse transcriptase conserved sequences included transition, transversion, deletion, and mutation sites showing preference. The base mutation frequency implanted by low-energy N+ was nearly 2-fold compared to Co-60 gamma-ray irradiation. Amino acid sequence analysis showed that deletion mutation and stop codon mutation caused by low-energy N+ implantation may be the main reason for the retrotransposons heterogeneity. Learned the sensitivity and mutagenesis of WIS2-1A to different physical radiation source provided the basis for theoretical study of biological organisms to the mutagenic effects.
