Simultaneous determination of trans- and cis-resveratrol in spiked plasma by high-performance liquid chromatography with photo-diode array UV-Vis and fluorimetric detection

摘要
An HPLC method has been developed for detection of both trans- and cis-resveratrol, extracted from plasma, by use of two detectors (photo-diode array UV-Vis and fluorimetric) connected in series. The isomers were separated by gradient elution on an octyl column with acetonitrile-water mobile phases delivered 1 mt min(-1) (run rime ca 30 min). The detection limit of the method was 2.5-10 ng mL(-1) and the quantitation limit was 5-25 ng mL(-1) plasma (depending on the detector used) with recovery ca 70 %. Linearity of response was ascertained in the range 5-1000 ng mL(-1). Short-term method validation was performed by intra- and inter-day assays at three concentration levels. All results satisfied predefined acceptance criteria.
