Scale-up of submerged culture Conditions for the Production of fibrinolytic enzyme with Cordyceps militaris

摘要
Cordyceps militaris is one of the medicinal mushrooms. A kind of enzyme with stronger fibrinolytic activity was found in the fermentation culture fluids of Cordyceps militaris. The fermentation conditions for the fibrinolytic enzyme production with Cordyceps militaris were investigated. And the production process was scaled-up from 250 mL flask to 10L fermentor. The results showed that the best medium for the fibrinolytic enzyme production was composed of sucrose of 2 % and bean cake of 5 %. The better inoculums size was punching out 1cm(2) mycelia cultures into 50 mL culture liquid of 250 mL flask. Liquid spawn of 0.5 % (v/v) was also better. The fermentation of 10 L fermentor under the conditions of agitation rate 100 r/min and aeration rate 600 L/h got a better result for the fibrinolytic enzyme production. Fibrinolytic activity reached 214.28 mm(2)/mL, equal to 286.21 U/mL of urokinase, which was 3.2 times more than the results of flask. The Y-P/x, was 5.5 times more than the results of flask.
