Simultaneous Determination of Three Flavonoids in Rat Plasma of Masson Pinus Needles (Pinus massoniana L.) Extracts by UHPLC-MS/MS and Application to Pharmacokinetics Studies after Oral and Transdermal Administration

摘要
Pinusmassoniananeedles, atraditionalherb, wereappliedtopreventhairlossinChina. Studiesavailablemainlyfocusedonpineneedleflavonoidswithvariousbiologicalactivities. However, therehasbeennopharmacokineticsstudyoftheflavonoidsfromPinusneedlesextract. Aselectiveandsensitiveultra-highperformanceliquidchromatography-tandemmassspectrometry(UHPLC-MS/MS) methodwasdevelopedtosimultaneouslyquantifytaxifolin, quercetinandcatechininratplasma. Toseparatethethreeconstituents, anAgilentExtendC18column(2.1mmx100mm, 1.8 mu m) wasusedwithamobilephraseof(A) 0.1% formicacidand(B) acetonitrile. Theanalytesweremeasuredbymultiplereactionmonitoringinthenegativeionizationmode. TherewasgoodlinearityintheestablishedUHPLCMS/MSmethod, withacoefficientofdetermination(r(2)) of>0.99. Theaccuracy, intra-dayandinter-dayprecisionandrecoverywereallsatisfactoryandthese3compoundswerestableunderthetestedconditions. ThevalidatedmethodinthisstudywassuccessfullyappliedtopharmacokineticstudyinhealthyratsafteroralandtransdermaladministrationofPinusneedlesextract.Theresultscouldprovidefurtherresearchfoundationforpineneedleextractasexternalpreparations.
