LPS、TNF-α、IL-1β刺激RAW264.7细胞分泌NO的比较

摘要
目的研究LPS、TNF-α、IL-1β刺激RAW264.7细胞分泌NO的剂量、时间依赖性关系,为建立炎症细胞模型提供实验依据。方法以LPS(0.01、0.10、1.00、10.00μg/m L)、TNF-α(0.10、1.00、10.00、100.00 ng/m L)或IL-1β(0.10、1.00、10.00、100.00 ng/m L)刺激RAW264.7细胞,24 h后收集细胞上清液Greiss法测定NO的含量。以LPS(1μg/m L)、TNF-α(10 ng/m L)或IL-1β(10 ng/m L)分别刺激RAW264.7细胞6、12、24、48 h,收集细胞上清液Greiss法测定NO的含量。结果不同剂量LPS均可显著诱导RAW264.7细胞分泌大量NO,无明显的剂量依赖性,不同剂量TNF-α或IL-1β均可显著诱导RAW264.7细胞分泌NO,且有一定的剂量依赖性,LPS(1μg/m L)或TNF-α(10 ng/m L)刺激RAW264.7细胞12~48 h时NO的分泌量显著升高,IL-1β(10 ng/m L)刺激6~48 h均可显著诱NO的分泌,均具有一定的剂量依赖性。结论0.01~10.00μg/m L LPS、0.10~100.00 ng/m L的TNF-α或IL-1β刺激RAW264.7细胞均可成功复制炎症细胞模型。
