miR26a-1 rs7372209和miR-612 rs12803915位点多态性与海南地区肺癌遗传易感性的关联研究

摘要
目的:探讨海南地区人群miR26a-1(rs7372209,C>T)及miR-612(rs12803915,G>A)多态性是否与肺癌遗传易感性相关,为本地区人群肺癌的防治提供理论依据。方法:采用病例-对照的研究方法,征得符合纳入标准的研究对象同意后,对其进行问卷调查并采集外周静脉血,提取DNA,采用SNaPshot技术进行基因分型,利用卡方检验、Logistic回归分析等统计学方法分析两位点多态性与肺癌发生的关联强度。结果:本次调查共纳入病例组142名,对照组211名,两组年龄、性别及肿瘤家族史构成无差异(P=0.258、0.143、0.688),病例组的吸烟率和饮酒率均显著高于对照组(P=0.002、0.023);校正混杂因素后,rs7372209位点突变等位基因T携带者(即CT和TT基因型)较携带野生基因型CC者的肺癌发生风险降低42%(OR=0.58,95%CI=0.37~0.90,P=0.016),并且这种保护作用关联性在不吸烟人群(OR=0.50,95%CI=0.26~0.96,P=0.038)及不饮酒人群(OR=0.52,95%CI=0.31~0.86,P=0.011)中更为显著。结论:miR26a-1rs7372209位点T等位基因降低了海南地区人群,尤其是非吸烟和非饮酒人群的肺癌发生风险。
