Preparation of phospholipid-docosahexaenoic acid catalyzed by immobilized phospholipase A1

A facile enzymatic synthesis approaching to prepare phospholipid-docosahexaenoic acid (PL-DHA) has been investigated through the phospholipase A1 (PLA1)-catalyzed reaction of docosahexaenoic acid (DHA) as well as phospholipid enriched in phosphatidylcholine (PC). The alteration elements of different parameters such as molar ratio of substrates, reaction temperature, enzyme dosage, solvent dosage, water activity and reaction time have been researched systematically. The ratio of DHA and PC was 4:1(mol/mol), the enzyme dosage was the 20% of two substrates, the solvent dosage was 4 mL and the reaction temperature and reaction time were 40 and 2 h. By the quantitative analysis of GC, the content of DHA was detected to be 17.7 mg/g PL on the optimal condition. The variation of PC content before and after the reaction was also analyzed by The Thin Layer Chromatography-Flame Ion Detector (TLC-FID). ©, 2015, Editorial Department, Chinese Cereals and Oils Association.
