Localized surface plasmon resonance (LSPR) based nanobiosensor in order to human growth hormone detection

Human growth hormone (hGH) regulates a number of metabolic processes involved in children growth and is medically prescribed to treat some diseases such as growth disorders. So, the hGH detection of serum is of great importance. Novel biosensors seem to be more advantageous than traditional techniques which are difficult and expensive. Accordingly, designing a rapid and highly sensitive localized surface plasmon resonance (LSPR) biosensor for hGH detection is the aim of this study. Herein, rabbit polyclonal anti-hGH was produced following 12 weeks of immunization by hGH injection and anti-hGH purified using protein G affinity chromatography. In addition, the spherical gold nanoparticles (GNPs) were synthesized by sodium citrate reduction of HAuCl 4 . Nanobiosensor was designed based on electrostatically conjugation between the antibody and GNPs. The conjugation has been confirmed with dynamic light scattering (DLS) technique and the sensitivity of nanobiosensor was studied via tracing the LSPR band Λ max shifts of GNPs. Ultimately LSPR sensitivity of nanobiosensor was compared to indirect ELIS A results. The data showed that the sensitivity of LSPR nanobiosensor and ELISA is 5 ng/ml and 5 μg/ml, respectively. These results indicated that LSPR nanobiosensor has the potential to be applied for the rapid detection of hGH instead of the traditional methods.
