MiRNA-27a通过PPARγ介导3T3-L1细胞胰岛素抵抗

摘要
目的初步探讨miRNA-27a介导3T3-L1细胞胰岛素抵抗的机制。方法通过肿瘤坏死因子α(tumor necrosis factor-alpha,TNF-α)诱导建立3T3-L1脂肪细胞胰岛素抵抗的细胞模型,观察miRNA-27a在3T3-L1细胞胰岛素抵抗模型中的表达变化,逆转录聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)检测miRNA-27a对过氧化物酶体增殖物激活受体γ(peroxisome proliferator activated receptor-gamma,PPARγ)mRNA的调控作用。结果miRNA-27a在3T3-L1细胞胰岛素抵抗模型中表达上调,miRNA-27a模拟物抑制PPARγmRNA的表达,miRNA-27a抑制剂上调PPARγmRNA的表达。结论 miRNA-27a介导的3T3-L1细胞胰岛素抵抗可能是通过靶向PPARγ实现。
