EVIDENCE IN SUPPORT OF THE CAMPBELL MECHANISM FOR THE PRECISE EXCISION OF COMPOSITE TN9 TRANSPOSONS IN ESCHERICHIA-COLI-K-12

摘要
The slippage mechanism envisages the precise excision of the composite transposons Tn5 and Tn10 (inverted IS elements) as an error in the replication of DNA. We have shown that, at least for the composite Tn9 type transposons (IS elements in direct orientation), precise excision proceeded by the Campbell mechanism through recombination along direct short repeats. A transposon containing the region of replication initiation was constructed on the basis of Tn9. Following events of precise excision we found circular transposon structures containing a direct short repeat at the junction of the IS elements.
